R.B.I Baseball 16's main interface has a straightforward design. The color scheme is easy to see without complicated patterns. The interface looks like a stadium's outer rim with a curved shape. The baseball field itself is shaped like a fan, which is similar to the designs of real stadiums. When a goal is scored, the game's score and animations of cheering fans appear behind the game. The score and animations appear to the right of the game with a control button below it for players to press.

When preparing for your game, you need to find a team first. Each team needs nine players; two sides are formed from this. One side must have a center fielder who plays the correct position. The remaining players fill the defensive positions. When the pitcher delivers the ball, the attacking team must hit it. Nine of the team provides nine opportunities to change each time the current batter misses. When the opposing team catches the ball in the air, but hasn’t touched the ground, they become “out” for the current round. The next batter has to be someone from the team who replaces them— this is the general rule governing all teams. The main command buttons rotate and hit the ball, which players must use. They can also run the map and navigate the options available at the bottom of the screen. Once players understand these commands, they can decide if their hand is fast enough to play.
